ZENKER's Fixative
Zenker fixing solution is a special fixing solution used in histology and pathology to prepare tissue specimens for microscopic examination. It consists of a mixture of mercuric chloride (HgCl2), potassium dichromate (K2Cr2O7), and sodium sulfate (Na2SO4).
The main function of Zenker Fixing Solution is to stabilize tissue structures and cellular components and maintain their morphology during further processing and staining. The solution works by a combination of cross-linking, precipitation, and coagulation of proteins and other cellular components to provide a consistent structure for microscopic analysis.
An advantage of Zenker Fixing Solution is its ability to provide excellent tissue preservation and cell structure detail, especially for the study of nucleic acids and nucleoproteins. It is particularly useful for staining chromatin and nucleoli in cell nuclei, as well as for imaging fibrils and other fine structures.
However, the use of mercuric chloride in Zenker Fixing Solution is associated with certain safety concerns and disposal requirements. Mercury is toxic and harmful to the environment, so precautions should be taken and appropriate disposal methods used when working with this solution.
