Staining Solution for Leukocytes
Leukocyte staining solution, consisting of a special solution of phloxin B and benzidine in 50% ethanol, is an alternative method for visualizing white blood cells (leukocytes) in blood or bone marrow preparations.
Phloxin B is a synthetic dye that can selectively stain cell structures such as nuclei and cytoplasmic granules. Benzidine, an organic reagent, serves as an oxidizing agent that improves the staining efficiency of phloxin B and reduces background staining. The combination of phloxin B and benzidine in 50% ethanol allows differential visualization of the different types of leukocytes, such as lymphocytes, neutrophils, eosinophils, basophils and monocytes, based on their morphological characteristics, nuclear shapes and granularity.
