Staining Kit: HEROVICI for collagen differentiation
The reactions in the staining process are based on the formation of metal complexes that selectively stain the collagen. In this process, the metal ions from the iron alum solution and the aluminum-iron hematoxylin bind to the collagen fibers and generate color options for their differentiation. The metanil yellow stains mature collagen fibers yellow, while the coelestin blue stains young collagen fibers blue. Acetic acid facilitates staining and serves to regulate pH, while lithium carbonate neutralizes the staining solutions. Picropolychrome solution contributes to differential staining by allowing further nuances.
Differential staining of collagen fibers allows detailed analysis of tissue samples, which may be important for medical diagnostics or research purposes. Successfully performed Herovici staining can reveal differences in the structure and composition of tissues and possible pathological changes.
In summary, the staining kit is used for differential collagen analysis.

Instructions / Protocol / Recommendations
Verwendung:
The HEROVICI staining kit is a product for in vitro diagnostics and is used for collagen differentiation in histological specimens. The combination of coelestin blue iron alum solution, aluminum iron hematoxylin, metanil yellow for Heravici stain, acetic acid 1%, lithium carbonate 0.05% and picropolychrome solution allows special staining of younger collagen in blue color and older collagen in red color. In addition, this staining helps to visualize reticulin in blue and cytoplasm in yellow. HEROVICI staining is particularly suitable for visualizing processes of wound healing, as it visualizes the healing process and maturation of collagen.
Prinzip:
Herovici staining is a histological staining method for the differentiation of collagen fibers in tissue or cell structures. The staining is based on the binding of aluminum-iron-hematoxylin and coelestine blue-iron alum solution to collagen substances and is modulated by the interaction with metanil yellow, acetic acid and lithium carbonate. Picropolychrome solution is used to remove unbound dyes. In this stain, young collagen fibers give a bluish color, while mature collagen fibers take on reddish-brown tones. This enables the detailed examination of collagen structures and the analysis of inflammation, ageing and degenerative processes in in-vitro diagnostics.
