Eosin 1% alcoholic 70%, acidified 0.5% Acetic acid
The use of 70% ethanol has several advantages, as it allows for faster staining while providing better fixation of cells during the staining process. The addition of 0.5% acetic acid in the solution has two main functions. First, it allows better diffusion of the dye into the tissue, resulting in more uniform staining. Second, the acetic acid promotes the decolorization of excess eosin, resulting in improved differentiation of cellular and extracellular structures.
Eosin 1% alcoholic 70%, acetic acid 0.5% is therefore a good choice for histological and cytological applications where rapid and uniform staining and improved differentiation of cellular and extracellular structures are important. The use of ethanol and acetic acid provides optimal staining that preserves the morphological characteristics of cells and tissues and allows adequate differentiation of cellular and extracellular structures.

Verwendung:
Eosin staining solution is used to counterstain proteins, connective tissue, fibres and keratin after core staining with haematoxilin. Smear preparations, frozen sections, thin-layer preparations or paraffin sections are used for staining. The staining solution is intended exclusively for professional use as an in-vitro diagnostic agent in histological or cytological laboratories. 1% Eosin staining solution, alcoholic can be used individually or in combination with haematoxylin (e.g. item no.: 10231) in H&E staining.

Eosin (tetrabromo-flourescein sodium) is stained regressively, i.e. it is first overstained and then differentiated. The eosin present in acidic solution stains cytoplasm and extracytoplasmic structures (collagen). The release of H+ ions from the acidic solution gives the cytoplasm a positive charge. Negatively charged eosin can bind to the cytoplasm. Eosin staining can be intensified by pre-treating the tissue with acid or adding a few drops of acetic acid to the staining solution. This leads to an increased accumulation of H+ ions on the amino groups of the proteins. A pH value between 4 and 6 is usually set. The pH value of the solution influences the colour shade. With alcoholic solutions, the same effect can be achieved (but without the possibility of specifying a pH value) by adding defined, empirically determined quantities of acetic acid to the alcohol. solution to achieve the desired result.
