Verwendung:
Hydrochloric acid alcohol (3 % / 70 %) is used in in-vitro diagnostics, especially in histology. The solution is primarily used as a differentiating agent for (iron) hematoxylin staining (regressive method). It selectively removes excess haematoxylin from tissue sections so that the cell nuclei are clearly violet. In addition, this mixture is used to decolorize tissue layers that have already been stained, resulting in complete decolorization. This allows the tissue sections to be re-stained for different diagnostic examinations, thus increasing flexibility in histological analysis and optimizing the diagnostic value of tissue samples.

In hematoxylin staining, the solution described acts as a differentiating agent that helps to remove excess or non-specifically bound dyes from the tissue samples in order to highlight the specific staining of certain cell and tissue structures. For this purpose, the hydrochloric acid in the solution selectively attacks the bases in the tissues, resulting in the loosening and partial removal of the dye, while the alcohol acts as a solvent that absorbs the dyes and dissolves them out of the tissues. This selective decolorization makes it possible to visualize structures such as cell nuclei and cell plasmas in different color intensities, thus enabling a clearer and more precise microscopic assessment and diagnosis in pathology.
