Picric Acid, saturated aqueous

Picric acid is a yellowish chemical compound used in histology as a fixative. An aqueous saturated picric acid solution consists of picric acid dissolved in water until no more picric acid can be dissolved in the solution. The saturated solution has a yellowish color and is often used in combination with other fixatives such as formalin or glutaraldehyde.
In histology, picric acid is used to fix tissue specimens to preserve their structure and morphology during processing and staining. Picric acid has a special affinity for basic proteins such as histones, which are found in cell nuclei, and thus can preserve nuclear structures in cells well. It is also useful for fixing cell structures with high lipid content because it is lipid soluble and does not tend to extract lipids from tissues.
Picric acid is explosive when dry and under extremely high mechanical stress. However, when an aqueous saturated solution is used, the risk of explosion is significantly reduced. Nevertheless, care should be taken to prevent the solution from drying and coming into contact with metals to avoid the formation of sensitive picrates.

Picric acid is also used in metallography, for the etching of certain metals and alloys. Etching is an important step in metallographic sample preparation to reveal the microstructure of metals and alloys. It involves selectively attacking the surface of the sample to highlight grain and phase boundaries and other microscopic features.

The aqueous saturated picric acid solution is used in metallography primarily for etching stainless steels, nickel-based alloys and certain aluminum alloys. By using picric acid as an etchant, the different phases and grain structures of these materials can be visualized and analyzed. This makes it possible to draw conclusions about the mechanical properties and behavior of the material under different conditions.

In metallography, the picric acid solution is usually used as part of an etchant mixture that may contain other acids or chemical compounds to optimize etching performance and selectivity.
