Periodic Acid 3 %
Periodic acid 3% is an aqueous solution of periodic acid (HIO4) used in histology and cytology for various applications. Periodic acid is a strong oxidizing agent and has the ability to oxidize complex carbohydrates and glycoproteins in cell and tissue samples.
One of the main uses of Periodic Acid 3% in histology is to perform periodic acid-Schiffs reaction (PAS reaction). This method is commonly used to visualize polysaccharides, glycoproteins, glycolipids, and other carbohydrate-containing structures in tissue samples. In this technique, periodic acid followed by Schiff's reagent oxidize carbohydrates in cellular structures, producing a characteristic purple to magenta color.
The PAS reaction is particularly useful for visualizing glycogen, mucus, basement membranes, and other carbohydrate-containing structures in various tissue types. This technique has applications in the diagnosis of various diseases, in the study of tissue organization and structure, and in the study of carbohydrate-containing substances in biological samples.
