Paraformaldehyde (PFA) 4 %, in PBS pH 7.2
Paraformaldehyde (PFA) 4% in PBS (phosphate-buffered saline) with a pH of 7.2 is a commonly used fixing solution in histology and cell biology. PFA is a polymer of formaldehyde and is depolymerized to formaldehyde by heating in aqueous solution. The 4% PFA solution in PBS is used to fix cells and tissues by cross-linking proteins and nucleic acids, thereby preserving cellular structure and morphology.
The advantage of using PFA in PBS solution is that the pH and ionic strength of the environment are stabilized during fixation, which helps to better preserve the natural properties of the material being fixed.
