Papanicolaou’s Solution - EA31 (PAP 3a)
Papanicolaous solution EA31 (PAP 3a) is one of the staining components used in Papanicolaou staining (PAP staining). PAP staining is a multi-step staining technique used primarily in the cytological examination of cells in body fluids, especially for cervical smears in gynecological cytology.
The EA31 solution is a mixture of eosin Y, light green, and bismark brown. It is used in the third step of PAP staining after the nuclei have been stained with Gill's Papanicolaous Hematoxylin (PAP 1b). The EA31 solution stains the cytoplasm and various cytoplasmic structures, with orangephilic and eosinophilic components taking on different colors. The resulting differential staining allows detailed examination of cell morphology and facilitates diagnosis of cell changes or abnormalities.
