PIANESE’s Staining Solution
The PIANESE staining solution is especially suitable for the differentiation of fungus-infected plant material in kerosene sections. With its application, host cells can be shown green and fungal mycelium bright pink. By treating with sodium hypochlorite/acetic acid before staining, cell contents can be removed to achieve better contrast.
When used, the staining solution shows a clear separation between fungal and plant cells. However, performance may vary depending on section thickness. Thicker sections (15-20 microns) contain more mycelium and therefore can be stained more strongly, although they also contain more cells and possibly misstained structures. Thin sections (5-10 microns) stain more weakly, but allow for better detection of mycelium.
In addition to its use in identifying fungal infections, PIANESE staining solution is also useful for visualizing trichomes and highlighting lignified structures that may occur in response to infection.
