Kit: Developer Solution for WARTHIN-STARRY Silver Staining

The Warthin-Starry Silver Staining Developer Solution Kit is specifically designed for the histological detection of Helicobacter pylori, spirochetes and other Gram-negative bacteria. Silver staining using the Warthin-Starry technique allows the examination of thin tissue sections and provides high sensitivity and specificity for the detection of these microorganisms.

The kit's special suitability lies in its ability to visualize bacteria in complex tissue samples that are difficult to detect using conventional staining techniques. The gelatin solution, hydroquinone solution and silver nitrate solution together form the staining system that selectively stains bacteria and enables their identification.

The kit's chemical mode of operation is based on the reduction of silver nitrate by hydroquinone in the presence of gelatin. The reduced silver ions form dark silver particles that bind to the bacteria and make them visible under the microscope. The gelatin solution ensures uniform distribution of the silver particles and facilitates binding to the target structures.

Verwendung:

The Warthin-Starry stain is used for the histological detection of Heliobacter pylori in tissue samples using light microscopy. Various modifications of this stain are used for the targeted detection of other bacteria, such as Bacillus piliformis and spirochetes. Detection is usually carried out in samples of the surface epithelium of the gastric glands and gastric mucosa.

The Warthin-Stary stain is a classic silver nitrate stain in which silver nitrate is reduced to metallic silver with the aid of a hydroquinone-containing developer. The reaction is stopped after a certain time by a rinsing step. As a result, the tissue is stained yellow-brown to golden-brown, while bacteria stand out in black. Cell nuclei take on a dark brown colour. The staining is stabilised by a final fixation reaction in sodium thiosulphate. If this step is not carried out, the sections will bleach out after some time.
