Hematoxylin 10%, alcoholic is an effective solution widely used in histology, in vitro diagnostic and scientific laboratories. The solution is mainly composed of ethanol 99.0% denatured (CAS No.: 64-17-5) and hematoxylin (C.I.: 75290) (CAS No.: 517-28-2, 475-25-2). Hematoxylin is a natural dye derived from the heartwood of various tree species and has the ability to stain acidic structures in cells, especially nuclei, DNA, and rough endoplasmic reticulum, blue.
The chemical formula of hematoxylin is C_16H_14O_6. In solution, hematoxylin and metal salts, usually aluminum, react to form hematoxylin-aluminum complexes, which are responsible for the blue coloration. This results in a so-called oxidation of the phenolic hydroxyl groups of hematoxylin, giving them a basophilic property.
Blue staining of basophilic structures allows visualization of cell nuclei and other fine structures to facilitate examination of cell tissue samples. This is particularly useful in disease diagnosis, cell biology research, and tissue sample analysis.
