Formalin 4.5%, carbonate buffer, pH neutral consists of calcium carbonate (marble) (CAS No.: 471-34-1), formaldehyde stabilized 37% (CAS No.: 50-00-0 / 67-56-1) and distilled aqua. / deionized water (CAS No.: 7732-18-5). It is a ready-to-use solution used as a routine fixative in histology. The solution is suitable for in vitro diagnostics and finds application in scientific laboratories.
Chemically, the solution consists of formaldehyde (HCHO), an aldehyde that can disproportionate in water to methanol (CH3OH) and formic acid (HCOOH). Calcium carbonate (CaCO3) serves as a buffer to maintain the pH of the solution at about 7.0. The buffering capacity is achieved by the reaction of calcium carbonate with water and carbon dioxide (CO2), forming calcium hydrogen carbonate [Ca(HCO3)2].
Fixation of tissue samples is achieved by the reaction of formaldehyde with proteins, especially amino groups, forming methylene bridges (CH2). This reaction stabilizes cell structures and allows detailed study of morphology and cell architecture.

Bei der Fixierung werden alle in der Zelle ablaufenden Prozesse unterbrochen, unter möglichst optimaler Erhaltung des Zustandes und der Struktur des Gewebes. Biochemische Prozesse wie Autolyse, Verwesung und Fäulnis werden wirkungsvoll verhindert. Fixierlösung nach Stieve eignet sich zur Fixierung von Hodengewebe und Eierstockgewebe. Die Fixierlösung ist ausschließlich für die professionelle Anwendung als in-vitro Diagnostikum im histologischen Labor vorgesehen.
