Staining Kit: ROQUE’s Trichrome Staining (CAB)

Trichrome according to ROQUE (CAB) is a histological staining kit consisting of four essential components: WEIGERT Stock Solution A, Phosphomolybdic Acid 1%, Chromotrope Aniline Blue Solution and WEIGERT Stock Solution B. The product is specifically designed for visualization of collagenous connective tissue (blue) with differentiation of cytoplasm and muscle tissue (red). It is particularly used for the detection of Mallory bodies in liver tissue.
The chemical reactions and properties of the product are based on the use of acid dyes and polyacids. Normally, acid dyes would stain the same basic proteins, but sequential application can manipulate the staining pattern. A polyacid, such as phosphomolybdic acid, is used to selectively remove dye. Polyacids behave like high molecular weight dyes: they displace easily removable dye from collagen.
The application of this staining kit can produce different results, depending on the specific application. For example, staining is used to differentiate muscle tissue from collagen, to distinguish pituitary alpha cells from beta cells, to identify increases in collagenous tissue such as in liver cirrhosis, and to distinguish tumors arising from muscle cells and fibroblasts.
