Staining Kit: TB Staining (cold) after ZIEHL-NEELSEN
The Ziehl-Neelsen TB staining kit (cold) is a specially developed staining kit for the detection of acid-fast mycobacteria such as Mycobacterium tuberculosis. Ziehl-Neelsen cold staining is an alternative method to Ziehl-Neelsen hot staining in the diagnosis of tuberculosis and other infections caused by acid-fast bacteria. Cold staining does not require heating of the specimen and is therefore advantageous in situations where a heat source is not available or heating is undesirable.
The staining kit contains the following components:
1. MucoFlutol (Item No.:12097): This solution is used to pre-treat the specimen to ensure optimal staining.
2. Ziehl-Neelsen carbol fuchsin (cold stain, Item No.:13070): This red dye allows good penetration into the cells. The sample is first treated with the cold fuchsin solution, with dye binding to acid-fast bacteria without heating.
3. Hydrochloric acid alcohol (3 % / 90 %, Item No.:12255): After treatment with cold fuchsin, the sample is treated with a hydrochloric acid alcohol solution to decolorize non-acid-fast cells and structures. Acid-fast bacteria retain the cold fuchsin dye and appear bright red in color, while other cell structures lose color.
4. Malachite Green Oxalate Solution (Item No.:12249): To improve visualization and differentiation of structures within the tissue sample, counterstain with malachite green oxalate. This solution stains the background and non-acid-fast cells a green hue, making the acid-fast bacteria easier to identify in contrast to other cellular structures.
