Staining Kit: NEISSER’s Staining after GIN
The staining kit: NEISSER staining according to GIN is a complex instrument for microbiological staining, which enables differentiated visualization of cells and cell structures. The included solutions I to III - methylene blue, crystal violet and chrysoidin - interact with specific chemical groups in cells and lead to selective staining.
The methylene blue solution (Neisser solution I) interacts with acidic structures and can thus visualize cellular components such as nucleic acids. Crystal violet (Neisser Solution II) has a high affinity for lipopolysaccharides in the cell wall of Gram-negative bacteria and stains them blue-violet. Chrysoidin (Neisser solution III) shows a preference for proteins and stains them yellow-orange.
Lugol's solution with lactic acid is an iodine-potassium iodide complex capable of recognizing starch and staining it dark blue. The lactic acid assists in the penetration of cell structures by the iodine solution, thus improving staining results.
Overall, the staining kit allows for differential visualization of cells and cell structures, and by combining the different dyes, a variety of information about the cells can be obtained, including their cell wall structure, protein content, and the presence of starch.
