Staining Kit: MOVAT’s Pentachrome Staining (Original)
The solutions included in the staining kit are:
1. Alcian blue 1%, in acetic acid 1% (pH 2.0): Stains mucus and proteoglycan-containing structures in blue.
2. Alkaline alcohol with ammonia (90/10): Used to pretreat tissue prior to hematoxylin staining.
3. WEIGERT stock solution A and WEIGERT stock solution B: Mixing the two stock solutions produces the Weigert iron hematoxylin solution, which is used to stain cell nuclei in black.
4. ferric chloride 2%: Used to differentiate the hematoxylin stain and to decolorize excess Alcian blue.
5. Brilliant-crocein acid fuchsin: Stains fibrinoid material and muscle tissue in red.
6. Phosphotungstic acid 5%: Used to decolorize excess Brilliant-Crocein acid fuchsin.
7. Safron du Gatinais: Staining collagen fibers in yellow.
8. Sodium thiosulfate 5%: Stabilizing the staining.
9. Acetic acid 1%: Used for differentiation and rinsing in the staining protocol.
Together, these components of the MOVAT Pentachrome (Original) staining kit enable specific staining and differential visualization of various tissue components with high differentiation of various hard and soft tissue components. Cartilage tissue is stained blue-green to dark green by metachromatic effects. Collagenous tissue (light yellow) can be differentiated from young osteoid (dark yellow), depending on fixation, osteon structures (reddish) are marked. Muscle tissue, cytoplasm and elastic fibers (red), as well as cell nuclei (blue-black) are differentiated.

Verwendung:
The MOVAT Pentachrome (Original) Staining Kit enables differential visualization of various tissue components, especially in cardiovascular and connective tissue structures, by combining five different colors in a single stain. It facilitates histopathological examination by allowing staining of mucus, collagen, elastin, cell nuclei and fibrinoid material. Applications range from imaging cartilage and bone tissue to differentiated imaging of hard and soft tissue components.

Alcian blue binds to acidic mucopolysaccharides, while alkaline alcohol with ammonia and WEIGERT stock solutions A and B provide staining of cell nuclei. Ferric chloride stains elastic fibers, while brilliant-crocein acid fuchsin and phosphotungstic acid are responsible for collagenous fibers. Saffron du Gatinais stains muscle fibers, and sodium thiosulfate and acetic acid are used for decolorization and solution optimization. This combination enables differentiated visualization of various tissue and cell structures in in vitro diagnostics.
