Staining Kit: KLEIHAUER’s HB-F Staining
The kit consists of the components KLEIHAUER stock solution A and B and KLEIHAUER erythrosine 0.1%, aqueous. When performing the Kleihauer-Betke test, stock solutions A & B are mixed together in a ratio of 5 : 1 (elution solution). The final counterstaining with erythrosine highlights the fetal cells containing HbF and allows clear differentiation from the maternal cells.
The KLEIHAUER HB-F stain provides a powerful tool for the analysis and quantification of fetal hemoglobin in maternal blood, aiding in the monitoring and management of pregnancies and fetomaternal blood transfusions.

Instructions / Protocol / Recommendations
Verwendung:
The KLEIHAUER HB-F staining kit is used for in vitro diagnostics of fetal cells in maternal blood by the detection of fetal hemoglobin (HbF). It allows the differentiation of maternal and fetal cells and assists in the monitoring and management of pregnancies, especially in cases of possible fetomaternal blood transfusion. The kit consists of two stock solutions (A and B) and one erythrosine solution, which in combination allow effective staining and analysis.
Prinzip:
The Kleihauer staining kit is a staining tool for the identification of hemoglobin F (HbF) in erythrocyte-containing tissues and cells in the context of in vitro diagnostics. This kit consists of three solutions: Stock Solution A, Stock Solution B, and Erythrosine. The solutions interact with each other and bind selectively to HbF in cells. This specific binding makes HbF visible in the cells, allowing differentiation between fetal and adult erythrocytes. This method has applications in the analysis of fetomaternal transfusion and the diagnosis of hemoglobinopathies by helping to determine the F content in hemoglobin.
