Staining Kit: FITE-FARACO’s Staining (Leprosy Detection)
Specially developed for the examination of tissue sections, this staining technique allows the identification of acid-fast mycobacteria in infected specimens. The stain is characterized by its high sensitivity and specificity, making it a valuable method in leprosy diagnosis.
The chemical operation of the FITE-FARACO stain is based on the use of carbolic fuchsin, a strong dye that penetrates the cell walls of mycobacteria and stains them red. Hydrochloric acid alcohol is used to remove excess carbolfuchsin and decolorize non-acid fast structures. Methylene blue serves as a counterstain that stains the surrounding tissue blue, providing contrast to the red-stained mycobacteria.
The advantage of this staining kit over other methods is its high sensitivity and specificity in identifying Mycobacterium leprae. The combination of the different components allows efficient staining and facilitates subsequent microscopic evaluation to make an accurate diagnosis.

Instructions / Protocol / Recommendations
Verwendung:
The FITE-FARACO staining kit is used for the detection of leprosy pathogens (Mycobacterium leprae) in histological specimens by in vitro diagnostics. It combines xylene peanut oil, Ziehl-Neelsen carbolfuchsin, 1% hydrochloric acid alcohol, and 0.25% aqueous methylene blue to identify acid-fast mycobacteria in infected tissue sections. The stain provides high sensitivity and specificity, making it a valuable method in leprosy diagnostics. The staining kit allows efficient staining and subsequent microscopic evaluation to make an accurate diagnosis.
Prinzip:
The FITE-FARACO staining kit is used for leprosy detection in in vitro diagnostics. It is based on a modified Ziehl-Neelsen staining procedure. In the procedure, samples are first treated with a fat-soluble dye (carbolfuchsin) that stains mycobacteria (leprosy pathogens). The sample is then differentiated with an acid-alcohol solution, which removes excess dye but allows the mycobacteria to retain their red staining. Finally, a contrast dye (methylene blue) is added to stain other cell structures blue, making the red mycobacteria more recognizable.
