CARNOY fixing solution (formaldehyde-alcohol-acetic acid)
CARNOY fixing solution is a histological fixing solution consisting of a mixture of formaldehyde, alcohol (usually ethanol) and acetic acid. It is used to preserve tissue specimens, maintain their structure, and prepare them for subsequent processing and examination under the microscope.
The CARNOY Fixing Solution has a strong fixing effect and offers several advantages over other fixing solutions. The combination of formaldehyde, alcohol and acetic acid results in rapid fixation of cell structures and prevents autolysis of cells. The solution quickly penetrates the tissue and allows efficient preservation of cell structures and morphology.
The alcohol component in CARNOY fixative solution allows simultaneous dehydration of the tissue, which benefits further processing steps. Acetic acid contributes to the fixation of nucleic acids and chromoproteins, resulting in improved visualization of cell nuclei.
The CARNOY Fixing Solution is particularly well suited for the fixation of chromosomes, as it allows for good morphology of cell nuclei and preserves chromosome structure well. However, due to its strong fixation properties and acetic acid component, it is not suitable for all staining and immunohistochemistry applications, as it may denature certain antigens and proteins.
When using CARNOY Fixing Solution, it is important to choose the correct contact time and ratios of components to achieve optimal tissue fixation and avoid possible artifacts or damage to the sample.
