BOUIN Fixing Solution

Bouin fixative solution is a histological fixative solution used in the preparation of tissue sections and cell preparations. It consists of saturated aqueous picric acid, formalin and acetic acid. This combination enables effective fixation and preservation of cell and tissue structures while preserving morphology and intracellular components. The solution also has a decalcifying effect by chelating Ca ions and lysing erythrocytes. Immunohistochemical imaging is possible with antigen masking.

The main use of Bouin fixative solution is in the examination of soft tissues such as testes, ovaries and intestines. It allows excellent preservation of cytoplasmic detail and visualization of extracellular matrix structures such as collagen fibers and reticulin fibers. Fixation with Bouin's solution is particularly advantageous for trichrome staining such as Masson's trichrome staining, as it allows sharp and differentiated visualization of the various tissue components.

The operation of Bouin's fixative solution is based on the chemical reactions of the individual components. Picric acid acts as a fixative that precipitates proteins and preserves cellular details. Formaldehyde stabilizes protein structures and preserves tissue morphology. Acetic acid aids in the fixation of cell nuclei and contributes to the preservation of cell integrity.

The special suitability of Bouin's fixative solution is in the study of tissues where the preservation of cell structures and morphology is important. Examples include reproductive tissues such as ovaries and testes, and inflammatory cells. For optimal fixation results, samples should be properly prepared and incubated in the solution for the appropriate amount of time. After fixation, specimens must be thoroughly washed and further processed to avoid unwanted fixation artifacts and to allow correct histological evaluation.

Verwendung:

During fixation, all processes taking place in the cell are interrupted, while preserving the condition and structure of the tissue as optimally as possible. Biochemical processes such as autolysis, decay and putrefaction are effectively prevented. Highly hydrous mesenchymal tissues and tissues with fine structures (e.g. testis) can be fixed very well with Bouin's solution. The fixing solution is intended exclusively for professional use as an in vitro diagnostic agent in the histology laboratory.
