Acridine orange 0.2 % with ammonia
Acridine orange 0.2% with ammonia consists of 1000 ml of aqua bidest / ultrapure water (CAS No.: 7732-18-5), 2 g of acridine orange (zinc chloride double salt) (C.I.: 46005) (CAS No.: 10127-02-3) and 3.3 ml of ammonia 25% (CAS No..: 1336-21-6) and is mainly used as an in vitro diagnostic agent, in histology and in scientific laboratories.
Chemically, acridine orange is a fluorescent dye with the formula C17H19N3-ZnCl2 and ammonia is an aqueous solution of NH3. The single solution exhibits fluorescent properties, which are caused by the presence of acridine orange in combination with ammonia.
The product is particularly suitable in histology for making cell structures in tissue samples visible by fluorescence microscopy. In this process, acridine orange selectively binds to nucleic acids of DNA and RNA, while ammonia adjusts the pH of the solution and thus influences the fluorescence properties.
The application of this solution enables the differentiation between living and dead cells, as acridine orange fluoresces green in living cells and red in dead cells. In addition, cellular structural changes such as apoptosis and necrosis can be detected.
