Paraformaldehyde (PFA) 10%
Paraformaldehyde (PFA) 10% is a highly concentrated fixing solution used in various fields of biology, especially histology and cell biology. PFA is a polymer of formaldehyde and is depolymerized to formaldehyde by heating in aqueous solution. The 10% PFA solution is used to fix cells and tissues by cross-linking proteins and nucleic acids, thereby preserving cellular structure and morphology.
The use of 10% PFA may in some cases result in greater cross-linking and better preservation of cellular structures, especially in thick tissue sections or larger tissue specimens that require greater fixation. However, a higher concentration of PFA may also result in greater cross-linking and increased hardness of the tissue, which may make it more difficult for antibodies or probes to penetrate during immunohistochemistry or in situ hybridization.
The 10% PFA solution is commonly used to fix tissues that are then embedded in kerosene or plastic to prepare them for microscopic examination. Care must be taken to adjust fixation time and conditions to achieve optimal fixation and tissue preservation.
