Formalin 4% with EDTA, pH 7.4, also known as Deicke's solution, is used as a fixing solution and preservative for biological specimens in histology, medical diagnostics and life sciences. It stabilizes cell structures and prevents degradation of specimens by inhibiting chemical reactions.

The solution is characterized by a particular suitability for the preservation of bone tissue, as the contained EDTA contributes to the demineralization of bone. This property allows better penetration of the fixative and thus improves the quality of histological sections.

The chemical properties of the solution are defined by the presence of formaldehyde and EDTA. Formaldehyde acts as a strong aldehyde that cross-links proteins and nucleic acids, thus fixing the structure of cells. EDTA acts as a chelating agent that binds calcium ions, contributing to the demineralization of hard tissue.

The functioning of the solution is based on the interaction between its chemical components and the sample. Formaldehyde reacts with the amino groups of proteins in the sample, forming a covalent bond that stabilizes protein structures. At the same time, EDTA removes calcium ions from the tissue, promoting its softness and penetrability.
