The Trichrome staining kit according to MILLIGAN offers a specific tool for in vitro diagnostics, which is particularly suitable for the visualization of muscle tissue and collagen in histological samples. Based on potassium dichromate hydrochloric acid, acid fuchsin, orange G bound to phosphomolybdic acid, free phosphomolybdic acid, acetic acid and Fastgreen, the kit allows differentiated visualization of various tissue components. The principle of staining is based on the selective binding of dyes to specific tissue components, with the mordants potassium dichromate hydrochloric acid and phosphomolybdic acid increasing the staining affinity of the dyes. Acid fuchsin and fastgreen interact with collagen and muscle fibers, while orange G targets cytoplasm and erythrocytes. Acetic acid acts as an acid fixative solution that regulates pH for optimal staining. By using the kit, muscle structures and collagen fibers can be visualized in tissue sections with high contrast sharpness and specific staining.
