Staining Kit: Picro-Sirius Red for Collagen I & III Detection

The Picro-Sirius Red staining kit is used for the visualization and differentiation of collagen type I and type III in histological preparations. It is particularly suitable for the analysis of connective tissue, supporting tissue, muscle tissue and cytoplasm in various organs and tissues.

The chemical mode of operation of this staining kit is based on the combination of Weigert stock solutions A and B, Picro-Sirius Red and acetic acid. Weigert stock solutions A and B are mixed in a 1:1 ratio just prior to use and are used for nucleus staining, while Picro-Sirius Red specifically binds to collagenous fibers and stains them red. Acetic acid is used as a solvent and to adjust the pH.

Compared to other staining methods, the Pikro-Siriusrot staining kit offers the advantage of not only visualizing collagenous fibers (red), but also differentiating muscle tissue and cytoplasm (yellow), in addition to providing nuclear staining. By examining the staining under polarized light, collagen type I (yellow-orange) and collagen type III (greenish) can be differentiated. This allows for more accurate analysis of tissue structure and collagenous connective tissue composition.

Overall, the Picro-Sirius Red staining kit provides accurate and specific staining of collagen type I and type III, improved differentiation of muscle tissue and cytoplasm, the ability to analyze collagen fibers under polarized light, and additional nuclear staining. Verwendung:

The Picro-Sirius Red staining kit for collagen is a special stain for visualizing collagen fibers (red) in connective and supporting tissue with differentiation of muscle tissue and cytoplasm (yellow) and cell nuclei (gray to black). In polarized light, this staining allows differentiation of collagen type 1 (yellow-orange) and collagen type 3 (greenish). Frozen sections, thin section preparations, and kerosene sections are used for staining. The staining solution is intended exclusively for professional use as an in vitro diagnostic agent in the histology laboratory.

Prinzip:

The basis of hemalaun staining is initially an oxidation of hematoxylin to hematein. Addition of metal salts (ferric chloride) leads to complexation of the metal ions with the hematein. A hematoxylin varnish is formed which is strongly positively charged. Staining is carried out in acidic solution. Nuclear staining with Weigert iron hematoxylin is particularly stable when counterstained with acidic dye solutions. Sirius red is an azo dye that binds to collagens due to its hydrophobic properties. Due to its large molecular structure, other tissue components are not stained. Due to its birefringence, collagens can be distinguished in polarized light.
