The NEISSER staining kit (diphtheria detection) consists of a combination of three NEISSER solutions: Methylene Blue, Crystal Violet and Chrysoidin. It is used in microbiology and bacteriology to detect Gram-positive pleomorphic rods such as Corynebacterium diphtheriae, the causative agent of diphtheria.

The NEISSER staining kit (diphtheria detection) is an in vitro diagnostic tool used in microbiology and bacteriology for the identification and diagnosis of diphtheria infections. Using a combination of methylene blue, crystal violet, and chrysoidin, Neisser staining allows the differentiation of morphological characteristics and granules in Corynebacterium diphtheriae, the Gram-positive, pleomorphic rod bacterium that causes diphtheria. Sequential application of these three solutions results in high-contrast, specific, and sensitive visualization of the bacteria under study and aids in the identification of diphtheria infections.

The NEISSER staining kit is used for the identification of diphtheria pathogens (Corynebacterium diphtheriae) in in vitro diagnostics. The principle is based on the selective staining of specific bacterial structures by the various dyes. Methylene blue stains predominantly the metachromatic granules, while crystal violet stains the cell walls of the bacteria. Chrysoidin is used for background staining to increase the contrast between bacteria and their surroundings during microscopic analysis. Due to the specific binding of the dyes to different structures, clear and effective visualization of diphtheria pathogens is possible.
