Staining Kit: Alcian Blue-PAS Staining
The special suitability of this kit lies in the differentiated visualization of tissue components. It allows differentiated analysis of tissue sections, especially in the diagnosis of diseases associated with altered composition or structure of connective tissue.
The chemical composition of the kit includes alcian blue, periodic acid, Schiff's reagent, and hematoxylin. Alcian blue binds to acidic mucosubstances and gives them a blue color. Periodic acid oxidizes neutral polysaccharides, which subsequently interact with Schiff's reagent and are colored red. Hematoxylin is used as a counterstain and produces the pink background.
The operation of the kit is based on the specific chemical interaction of the included reagents with the target structures in the tissue. This specific binding enables differential staining and thus visual differentiation of the various tissue components.

Alcian blue-PAS staining is a combination stain that uses the PAS reaction and the reaction of Alcian blue to visualize various carbohydrates depending on pH and electrolyte concentration. PAS staining is performed before staining with Alcian blue to stain neutral mucins, neutral glycoproteins and glycogen.\rAlcian blue PAS staining is particularly important in the diagnosis of lung tumors and lung metastases, as well as diseases of the gastrointestinal tract, but is also used for other carcinomas and disease patterns.
Prinzip:
Alcian blue is used in histology to selectively stain acid mucosubstances (acid mucins and sulfated glycosaminoglycans such as hyaluronic acid or chondroitin sulfate) at low pH (1.0 to 2.5). Depending on the pH of the staining solution, specific mucins can be detected. Another method of selective imaging, is to vary the electrolyte concentration. As the ionic strength increases, staining with Alcian blue is prevented, allowing differentiation of differently sulfated mucopolysaccharides as well as heparins, heparin sulfates and keratin sulfates.
