Crystal violet, a cationic dye, binds to cellulose in plant cell walls through electrostatic attraction. The resulting binding leads to a violet coloration of the cellulose structures. Auramine O, a fluorescent dye, finds application in the selective staining of lignin, a major component of secondary cell walls and lignified parts of plants. Auramine O is lipophilic and therefore binds to the lipophilic components of lignin, resulting in a yellow stain.

S SAFELINE Differentiation Solution and SAFELINE Rinsing Solutions are used to optimize the staining process by removing excess or unspecifically bound dye, while SAFELINE INCLUDAL PVA Solution facilitates sample fixation and preparation.
