Acetic Acid in Ethanol (1 % / 70 %)

The mixture of 1% acetic acid and 70% ethanol is mainly used as a differentiation step in histological staining. The main focus is on the differentiation of hematoxylin staining, e.g. in the context of hematoxylin-eosin staining (HE staining). In this process, it is used to remove excess hematoxylin, allowing precise staining of cell nuclear structures.

Another example is Papanicolaou staining (PAP staining), which is commonly used in cytology to identify abnormal cells. Here, the solution is used for differentiation and dehydration to achieve the desired staining results.

The solution is also an important component of trichrome staining, in which different tissue components are shown in different colors. In this case, it is used to remove excess dyes and produce a clear, differentiated image.
