Erylysis-Buffer pH 7.2 - 7.4
Erylysis buffer is an aqueous solution with a pH range of 7.2 to 7.4, consisting of a combination of potassium hydrogen carbonate, EDTA disodium salt dihydrate, ammonium chloride and ultrapure water.
The main uses of erylysis buffer are in hematology and cell biology, especially in the lysis of red blood cells (RBCs) and the isolation of leukocytes or other cell types. The buffer allows selective dissolution of red cell membranes and facilitates examination and analysis of remaining cells.
The chemical operation of the erylysis buffer is based on the hypotonic properties of the solution, which lead to the bursting of red cells, and chelation by EDTA disodium salt dihydrate, which inhibits the activity of proteases and other enzymes that could damage cell structures. Ammonium chloride and potassium bicarbonate contribute to the buffering capacity and osmotic balance required for the lysis step.
The special suitability of the erylytic buffer lies in its ability to selectively dissolve erythrocytes while maintaining the integrity of other cell types, such as leukocytes. This makes it an important choice for cell isolation and blood sample analysis in hematology and cell biology.
