GRAM’s Decolorizing Solution
Gram's decolorizing solution is an important component of Gram's stain, a classical microbiological staining method developed to classify bacteria into two main groups: Gram-positive and Gram-negative. The distinction is based on differences in the cell wall structure of the bacteria.
The destaining solution typically consists of a mixture of isopropanol and acetone. Destaining is a critical step in Gram staining because it removes the dyes from the Gram-negative bacteria while the Gram-positive bacteria retain their staining.
The Gram staining process consists of several steps:
1. Fixation of the specimen to the slide.
2. Staining with crystal violet solution, which stains both Gram-positive and Gram-negative bacteria.
3. Treatment with Lugol's solution (iodine solution), which acts as a mordant and enhances dye binding in bacterial cells.
4. Destaining with Gram's destaining solution. Gram-positive bacteria retain their purple color, while Gram-negative bacteria are decolorized.
5. Counterstain with a red dye solution, such as safranin. The decolorized Gram-negative bacteria take on this color and appear red, while the Gram-positive bacteria remain purple.
