BRADFORD reagent (5x concentrate) is a single solution used in medical diagnostics, histology and scientific laboratories. The product is particularly suitable for the determination of protein concentrations by the so-called Bradford method, a dye binding spectrophotometry method.

The solution consists of phosphoric acid (CAS no.: 7664-38-2), methanol (CAS no.: 67-56-1) and Coomassie Brilliant Blue G 250 (C.I.: 42655) (CAS No.: 6104-58-1).
The reaction that occurs when using the BRADFORD reagent is due to the interaction of the dye Coomassie Brilliant Blue G 250 with proteins. In the presence of proteins, the dye binds to them, shifting its absorption spectrum and developing a high absorption power in the region of 590-650 nm.

Chemical properties of the product result from the acidic solution provided by phosphoric acid and the solubility of the dye in methanol. During analysis, the amount of dye bound to proteins is determined by measuring the absorbance of the solution at a specific wavelength, which reveals the difference between bound and free dye.

Coomassie Brilliant Blue enters into a reaction with proteins, where it is indexed by the color change from brown to blue initiated by the proteins. Phosphoric acid provides the appropriate acidic pH in this process, while methanol acts as a solvent. The use of Coomassie Brilliant Blue benefits from its high affinity for proteins and its ability to form a strong Van der Waals bond.

The intensity of the color change can be used to draw conclusions about the concentration of proteins in the sample. This makes it possible to obtain an accurate overview of the protein content, allowing the reagent to be used, for example, in the diagnosis of diseases associated with altered protein levels.
