Alcian Blue 0.1 %

Alcian Blue 0.1% (pH 2.5 with acetic acid) is a histological staining solution mainly used for the detection of acidic mucopolysaccharides and acidic glycoproteins in tissue sections. The staining solution consists of 0.1% Alcian blue dissolved in a buffer with a pH of 2.5, prepared by adding acetic acid.

At this pH, Alcian blue binds selectively to acidic mucopolysaccharides and acidic glycoproteins, allowing differential staining of these structures. The stained substances appear blue, while the rest of the tissue remains largely colorless.

Alcian blue staining is widely used in histology and pathology to study the distribution and amount of acidic mucopolysaccharides and acidic glycoproteins in various tissues. This can be helpful in the diagnosis and study of diseases associated with changes in these substances, such as inflammatory diseases or tumors. Alcian blue staining is often performed in combination with other stains such as periodic acid-Schiff (PAS) staining or hematoxylin-eosin (H&E) staining to provide a more detailed picture of tissue structure and function.

Verwendung:

Alcianblau wird in der Histologie verwendet, um sauro Mukosubstanzen (saure Mucine und sulfatierte Glykosaminoglykane wie Hyaluronsäure oder Chondroitinsulfat) bei niedrigem pH-Wert (1,0 bis 2,5) selektiv anzufärben. Je nach pH-Wert der Färbelösung können spezifische Mucine nachgewiesen werden. Eine weitere Methode der selektiven Darstellung, ist die Variation der Elektrolytkonzentration. Mit steigender Ionenstärke wird eine Färbung mit Alcianblau verhindert, wodurch eine Differenzierung unterschiedlich stark sulfatierter Mucopolysaccharide sowie Heparine, Heparinsulfate und Keratinsulfate möglich ist.
